Every aspect of Prison School is part of a well-oiled machine that makes sure no part of the buffalo goes to waste. Jokes rarely stand on their own, they feed into further plot progression and characterization. If a joke appears to be nothing but gag existing in a vacuum (Andre's masochism, the principal's ass fetish, Chiyo's philosophy on people who like sumo) then it is only a matter of time until it pays off. The economy of comedy sustains the narrative and the character's relationships.
