Probably not, no. But I'd say taking things seriously usually ends up badly anyways, do you really want anime to become more mainstream.

Relatively mainstream audiences recognize the quality of things like Spirited Away and Akira, I'd count that as taking anime seriously
If you mean something along the lines of "Will people ever take me seriously for taking my Monster Musume dakimakura out on a walk in the park", then no, probably not

Japan takes anime seriously when it's good. Great Teacher Onizuka was adapted to a drama, and was the most watched tv show in japanese history at the time it aired. Your Name is breaking records, and even has bus tours to all the places shown in the film. There are more examples if you look. It's as seriously taken as something for teenagers/children possibly could (in its home country).
It's the equivalent of Harry Potter or Disney, I guess.

Yeah. I think the wonderful thing about anime is there are a lot of niche productions, so even as mainstream as some parts of it are in Japan, there's still the whole underside of the iceberg that remains as a, "special interest community".
