We've seen Bertolt be upset/hurt by Reiner's words, and they were during Castle Utgard when Reiner disassociated and saved Connie.

Basically Reiner wanted to boost Bertolt's confidence in one last bro talk before they got separated -- and as far as we know Bertolt doesn't have any hidden feelings about it. In fact he felt even more motivated by Reiner and wants to complete the mission for his sake.

I see the cave as worse because it's pretty much the only moment when he completely broke. His mom dying was without a doubt the worst loss he's ever suffered (though one could easily argue that seeing himself eat his own dad is just as horrible), but it's the loss that gave him the motivation to kill all the titans and strenghtened his resolve to free humanity from their caged lives. And for every subsequent loss he suffered, the culprits were always clearly defined.
The cave pretty much took all of that resolve and shat all over it. That's why I think of it as the worst moment for his character, cause he was made to believe that his own existence was going against everything he believed in and had been fighting up until that point.
