How did Rei even feel about Asuka? Did she reciprocate her hostility or did she just not give a fuck after Asuka refused her help?

Well she obsiously didn't like her, that's for shure. There where times where (pic related) she straight up confronts her on her bullshit. Most the time I say she was indiferent towards her, she probably saw her as a lost cause from the very beggining.

When they meet Rei has pretty much no opinion of Asuka, but as time goes on they grate on one another more and more. However, after the Matarael and Sahaquiel fights, Rei realizes that Asuka isn't really that horrible of a person and the two reach a bit of an understanding. After that though, everything goes to shit, and Asuka starts getting angry, hostile, and over-competitive and starts hating everyone around her, including Rei.
Asuka isn't specially hostile to Rei. That's how she behaves towards most people. And how many close friends Rei has? Exactly.
Just accept the main characters weren't really the people person kind. Rei would rather be alone reading and Asuka just wants to outdo the others or hang with Hikari. Speaking of which I always wondered why the class rep seemed to be the exception, who approached who and why they got along so well?
Could be. I mean at first she didn't event consider interacting with anyone but the other eva pilots. Until she actually meet the other two and realized it wouldn't work out.
But whatever the reason they actually got really close in a short time. Class rep would go to her for love advice, she slept over at Hikari when shit got bad.
Episode 12 is one of the best climaxes in all anime. You get to that point, and you think everything is going to work out well, but from episode 16 onward it's just one giant NOPE

Disliked her immediately, but not strongly enough to think of her much beyond "pilot of Unit 02", while also showing the most insight out of everyone but Misato into Asuka's persona and motivations.

Accurate. Rei treated Shinji pretty much the same to start, he manages to push through awkwardness and Asuka draws back at the first perceived sign of rejection. Not that Rei isn't still willing to join for ramen or hang out and watch lights on a hillside.

They're the most important girls in the series, even if there's other pairings that either make more sense (Asuka/Hikari) or are actually canon (Ritsuko/Maya)

Shinji scores pretty high on the Raglan scale. You have to make a few adjustments for the sci-fi anime setting but he's pretty by-the-numbers 18-22 scoring Raglan hero.
You know she would've hit that if everyone lived long enough though. Hyuga goes from "nice guy" doing her laundry to hacking into databases and other spy shit that Kaiji was doing when he was still alive, in order to help Misato get to the bottom of everything.
