Install the Intel INF update/Intel Chipset Device Software.
For some reason, the Intel INF update is necessary to tell Windows that the PCIe lane the xHCI controller is on has hot-pluggable devices attached to it. Without it, USB devices are only recognised immediately after booting.

I have a Z270 from MSI.
I reinstalled
-ASMedia USB3.0/3.1 Drivers
-Intel USB3.0 Driver
-Intel Chipset Driver
None of them helped.
Weird thing is, a few weeks ago it worked perfectly fine, but it has stopped working without me really changing anything at all on my machine.

Yes. It's a front port from the case.
All of them worked just fine a week ago or so, and now they suddenly don't. It can't really be a hardware fault though, USB 2.0 devices (e.g. XBox controller) work just fine on the 3.0 ports.

It certainly can be a hardware fault: USB 3 sockets have eight wires, and USB 2 socket only have four.

It's extremely unlikely though that both would just spontaneously die.
Plus, when I uninstall the stick's drivers, it'll come back to life on the first plugging in. Subsequent plugins either bug out or keep the stick dead.
