Although it's well documented (and pretty obvious) what the influences were for Rock (Mega Man)'s design, it's less clear for Roll. Astro Boy does have a sister, so the concept comes from there, but her design is radically different. Instead, I propose that it's Penny from Inspector Gadget that serves as the inspiration. Blond hair, blue eyed white girls, with red as the primary color.
It's not as far feteched as you'd think. The original Inspector Gadget series ran from 83-86. Mega Man was released in 87. Recently Japanese fans were asked to rank their top 20 Police Anime. Inspector Gadget ranked #12, showing it still has a cultural impact to this day.

Classic Roll's design is so generic that it's impossible to pin down any specific influences, but I think Francoise (from Cyborg 009) is one of them.
There are nine robot masters originally built by Dr. Light, just like there are nine original cyborgs in Cyborg 009.
And it seems not-so-coincidental that in both series, the girl is the third one created. Francoise is Cyborg 003, and Roll is DRN-002 (Blues and Rock being 000 and 001 respectively).
