This shit is popular because people have wanted Ash to have a steady girlfriend since the beginning of the series. They ship Ash with every female character, even minor ones.

Serena is the first main female protagonist that has a canonical crush on Ash.

But Ash is very clueless about romance; it's a recurring gag with him. So even though Ash sees Serena as a friend, he doesn't view her as a girlfriend or anything like that.

As for the chemistry, it was mostly Serena being influenced by Ash, which is an unhealthy dynamic.
