A tsundere is a character that displays cold, hostile behavior towards a certain character, while harboring kinder, gentler feelings beneath their surface.

In order for a character to fulfill the definition of a tsundere, they could have to display hostile characteristics towards a character, ranging from things like anger, annoyance, irritation, disgust among other things, for good reason or not, but at the same time, show their gentler side every now and then. Misty fulfills the definition pretty accurately based on her interactions with Ash, because while she's often hot headed in their interactions, she does have numerous moments where her dere side shows; notably in the Lavender Town episode where Ash technically dies, along with other episodes where she sympathizes with him when he loses a match, or releases a Pokemon.

It's also worth bearing in mind that Tsundere isn't simply an archetype, but a type of behavior. She behaves like a tsundere towards Ash, but towards every other character in the series, she's anything but, especially towards Pokemon not named Psyduck. To simply label her a "tsundere" or a "tsuntsun" is an extremely simplistic and narrow minded way of examining her character when, outside her interactions with Ash, she behaves like much more of a genki girl/deredere.

And that's just the tip of the iceberg; I'm sure what you're really wondering is what a "good, well-written tsundere" is. I can tell you right now that Misty isn't the first character that comes to mind in this regard, and as far as "well-written" characters go, you won't find any in Pokemon, at least if we're judging by the standards of genuinely good anime. Pokemon is a kids adventure show; the characters are very simple and that's all they need to be. If we're going to start comparing then to more complex examples of certain archetypes, there's always going to be a stark contrast in quality.

For anyone else who wants this explained further (not the retard I'm replying to), Even completely different archetypes can have occasional tsundere behavioral tendencies; Misty is primarily known for her tsundere behavior towards Ash, but yes, she does occasionally display them towards other characters, most notably her Psyduck, and sometimes (but rarely) Brock. If we're going to start classifying a character as a tsundere based on very sporadic scenes, then why stop at Misty? In the episode where Ash catches Bulbasaur, Brock displayed extreme tsundere behavior towards Misty; by this retard's logic, he must be a tsundere now.

People who actually watched the series realize whenever Brock gets infatuated with another girl, Misty's reaction is smugness/facepalming 95% of the time.

With May, she was sort of just going along with Ash to do the trainer stuff until she learned about contests and she decided to become a coordinator. With Serena, they had some episodes where they explore more of her strengths, such as the PokeVision stuff to put on a show and Poke Puffs competition before introducing the idea of Pokemon Performances, which ends up allowing Serena to make use of her previously shown strengths.
