Donkey Kong was a circus ape named after New Donk City, and his Kong family are named after the streets/landmarks. "Donkey Kong" the game took place in NDC.

Mario was born in the Mushroom Kingdom but somehow found his way to NDC as a boy (since they're connected), being raised as an Italian tradeworker. The incident in "Mario Bros." lead to Mario rediscovering the Mushroom Kingdom after following the Koopas through the pipes.
