I would say Jurassic Park, as long as it's during the summer. I went to Universal Orlando at one point in January, and they shut the waterfall off that the T-Rex is supposed to burst through at the end before the drop. It was kind of lame seeing it sit there lifelessly until it was cued up.
I don't care what anyone says, the Transformers and Harry Potter 3D rides are awesome too. Hell, if you like Harry Potter at all, it's fun just drinking some pumpkin juice, and taking in the atmosphere while you're waiting in line for the ride.

The Indiana Jones ride was the coolest shit when it first came out. The car bucked you around and shit. The whole ride was really dark so going through the dart tunnel all you'd feel is random puffs of air like they were whizzing right by your face. So fucking great. Waiting in line had videos with Sallah talking about the story for the ride and shit. I hear it totally sucks now though. They brightened it up big so there's no scary darkness and the car barely bounces you around taking away the feel of being in a real off road vehicle bouncing through an unstable temple.

This was originally planned as a ride dedicated to the movie Alien. The reason it appears so random and unrelated to anything is because its a workaround to the fact that they couldn't secure the concept of using the Alien franchise as a ride since it was rated R and was way too frightening for children. Imagine a fucking titular in that room, it would only be suitable for adults.
IMO Disney should have a park dedicated to adult themed R rated rides based off of the original concepts to this.
