Fat Dom disrespected the entire Soprano family by implying that they were homosexuals and making light of the death of Vito. Despite the Soprano family's homophobia, the death of one of its senior members at the hands of a rival family for any reason would have been a sore spot for years. Fat Dom should have known better than to press the issue while in Soprano territory. Accepting the disrespect would have made the Soprano family appear weak, encouraging further violence.
That said, Carlo and Silvio went too far in killing him, though their behavior isn't entirely unexpected given that most of the characters in the Sopranos are high-functioning psychopaths.
