What's a good method to lay the groundwork for a successful campaign?

Read fantasy/sci fi novels, history, and anything else that interests you and your players.
Steal liberally from all of them.
Start your players off with a small problem and let them work their way to the big problem you actually want them to solve. Horde of kobolds? They're running away from a witch who has control of a dragon. Farmer missing? An orc army is drafting slave labor.
Then you can pull together the disconnected mini-adventures you took your party on to get them warmed up to you and each other into a cohesive whole.
Finally: Bullshit liberally. The players often come up with better ideas for how the campaign should go then you do, so don't be afraid to let them speculate on what the end game is and steal that.

First things first, the players. Make sure your group is willing to play what you want to play. There's no "right" way to RPG, but if you don't make clear your intentions beforehand there will be conflict.
Second, make sure you know enough about the system to run it functionally. Know the Core Rule Book inside out. Be able to run a combat encounter without looking at anything but stat blocks. Its okay to not know corner cases, but have a solid grasp on how shit goes down in the game.
Finally, this is less groundwork, but be consistent and dedicated. Be sure you can run this game regularly, be able to take time out of your week to make sure you can prepare. Communicate with your players on how they're enjoying it. Don't get discouraged by a bad game or two, don't lose interest before everyone else.
