1000 Marines is the average size of a chapter. Despite the fan wankery that's not even enough men to crew a small warship let alone take over a planet. Sure, you could assassinate leaders, wreck bases, and topple governments. But you could never hold anything larger than a city. Hell, if you spread an entire chapter of marines across earth you would run out of marines before you ran out of cities to occupy!
You need someone to hold the territory. They don't need to be supermen, but they need to be able to enforce your will on the locals. Because the civilians of Glasgow may be cooperative when your in the same room as them, but lord knows what they're up once you take the train to London.
As for making more: You can't just train everyone to be supersoldiers. It's the third truth of SpecOps:
