What's our biggest problems with trying to colonize Mars?
Most people's immidiate answer will be getting there and I will partially agree with that but there are many, much more difficult things to deal with when going there.
First of all is that there is a very small window of time every year and a half (approx 19 months or so) where we can actually send rockets there because the earth travels around the sun at different speeds than mars. Because of this we would need to find ways to send enough resources for the people there to survive without help for potentially years at a time.
Next is the human body. We have evolved to be great at survivng in earth conditions. But being on mars' lower gravity will essentially kill us due to none decomposition and muscle decomposition. That along with how our circulatory system functions with the help of gravity means that humans won't be able to survive on mars without having a way to either simulate EarthLike gravity for extended periods or essentially blasting themselves with larger gravity that earth for shorter periods of time (most likely when sleeping) to help as a regulator.

Lol that was learned partly Elon musk, partly from Physics class, and partly from random sciences on how the human body works. The last part actually has a simpler answer than youd think. By sleeping in an elevator that has a way to move fast upwards but slow downwards it would simulate higher gravity on the upwards portion. Space suit is just so interesting to me.
