1. It's set on a Nintendo message board, this context sets up the joke. You're expecting a post that is both innocent and safe for work. The fact that the post is neither of these things is humorous.
2. The name and avatar of the poster are again, totally innocuous. It's this contrast with the content of the users post that accounts for a lot of why the image is so funny.
3. There's another user making a totally normal post below the first one. Once again reinforcing the humour of the above post being so out of place.
4. The post itself is written entirely in capitals. This jarring element along with the outrageous content of the post itself further assist with the humorous nature of the situation.
5. The post has three main elements that make it funny:
-The introduction, referring to people first as black and then as niggers is funny because typically it's either one or the other. Using both at the same time has a humorous effect.
-The act itself of attempting to police a group of people from posting is funny because we know the user holds no actual power in this regard. The act of someone totally powerless attempting to exert a level of control over something they have no control over is funny.
-The final threat of bringing back slavery isn't so much funny in itself but when paired with the context of the post and the users impotency regarding the matter concludes an already very funny image with another humorous note to go out on.
