Assuming the poison is potent enough to kill if you mix them together you can turn this into a log(N) problem by mixing them into 2 groups, see which kills and recursively divide and mix the barrels from the killmix until you end up with the poisoned barrel. Takes log(500)/log(2) ~= 9 dead guys.
