None of the characters have any real reason to get involved in a fighting tournament, or really do anything other than regular slice of life. Rainbow might enter, but she'd wimp out the moment she saw someone take a punch to the head. Pinkie might enter as a joke and just win by never getting hit.
I don't think there's any character's we've seen that would really be hyped at the chance to get into a fight though. Maybe Flash Sentry, if someone told him it would impress the ladies?

Twilight enters as well under a fake name, to gain experience in a field she has very little experience in. She makes it to the final.
Vinyl Scratch is also there, and she signed Octavia up without her permission, but it's too late for her to back out.
Big Mac is there for the prize money and wants to fix Granny's hip. Granny is also in the tournament.
Dash may or may not enter. It's a coin flip. On one hand, she wants to win. On the other hand, she has to fuck stallions.
Celestia wishes she could enter but can't, so she orders Cadence to instead.

Appul or Big Appul would win.

Pinkie doesn't have the desire to really slug it out and doesn't want to hurt her opponent. (Limey on the other hand might do better)

Dash is fast, if she can't but doesn't have the Endurance, unless she can take her opponent in the first few minutes, she's toast.

And the Appul's are stubborn, have endurance to go with it, and are stronk. They can take whatever Dash can dish out, and wait for her to tire out enough to put her on the ground.

Big Mac probably has the strength advantage over his sister, but AJ is more stubborn, and more competitive. Mac is more laid back. I'd give it to AJ.

It depends of what is allowed. If it's purely physicall contest without wings or magic probaby someone who is better fighter (I would choose Dash, she actually has some kind of training, or maybe Shing Armour, AJ maybe is stronger at least than Dash, but she isn't trained fighter).
If wings are allowed pegasus would destroy earth pony. Fighting with flying oponent when you are grounded is bad idea.
If magic is allowed too it's fifty fifty, if pegasus is fast enough to reach unicorn or can avoid getting shoot he or she has a chance to win but if unicorn can keep distance he is winner.
Also we seen many times that earth ponies aren't that much stronger or have bigger stamina than other pony kinds, beside their talent. If they aren't doing things linked to their cutie-mark they are not really that strong.
