Ahem, *if* you count Equestria Girls as canon, then it might be safe to assume that the ponies don't speak in any language that couldn't be replicated by a human throat—in other words, they don't speak through whinnies and neighs that have been translated into English. I say this because Twilight didn't freak out about any change in her voice or language when she went through the mirror, and she perfectly understood EqGmanoids. They must speak the same language in both worlds.
That's not to say they're speaking in a language we'd understand that's been translated, but they at least speak in a human-type language.

If EG is the case, then they speak a language that's close enough to English, but still a bit different (ie everyp0ny vs everybody). I say this because in one of the EQG books,I think it wasApplejack who referred to English as her favorite class in school. So EQG takes place on a different planet, in a strange country that happens to speak a dialect of English that's identical to ours

I like to think they have multiple written languages. At least three.
One is for printing. (the one that looks kinda like English)
Another that's the Equestrian equivalent to writing in cursive,(Gibberish one)
And a third that is probably short hand/hieroglyphic style (the one with the unicorn heads in it)

I'm going with that. The printing one is optimized for typing. The squiggle one is optimized for mouth/magic, and this one has minor variations that let the scribe leverage his or her dexterity. The hieroglyphics are for ancient or showy things.
The pure english one is just a translation so we get what's going on.
