Targaryens used to be fireproof, but now it's a lost trait--or was until Dany, either because a) she's a genetic throwback or b) magic has woken up in the world. Probably a combo of the two. Several Targaryens have burned trying to prove they had the right stuff.

Some Targaryens have immunity to fire but Dany's grandfather tried to burn himself with wildfire believing he would turn into a dragon. He did not have that immunity and thus burnt to death. Hope that clarifies.
