let's remove the obvious out of the way: if you go to Ciudad del Este don't spend your whole day in the shopping area, go further to the Republica Lake, grab a tour to Tati Yupi (http://www.companytour.com.br/turismo/foz-do-iguacu-tati-yupi-refugio), to the Itaipu dam, then take the bus back to Foz do Iguaçu since good hotels are only in Asunción and that's 305 kms from there
then you can go to Asunción for the nightlife (there isn't much to buy there except food and drinks, avoid the gift shops at the Microcentro) and Encarnación for the beach (by the river)
Villarrica has nice-ish nightlife too, 20 kms from there there's Oktoberfest in the middle of nowhere, a german colony called Independencia
as for Encarnación the nice season is february where we have our own Carnaval parade there. lots of girls (and guys if it's your thing)
and then.. it depends on what you want. Asunción is where the cumbia is at
