500,000 people is usually considered an important indicator of urban development. It usually means an area will have certain features of a "city" (larger hospitals, airports, cultural stuff, etc.). It also has implications for things like waste management and infrastructure.
A million or more people is usually considered a "large" metro area while 10+ million is a "megacity." Megacities of over 20 million are kind of in their own category.
