What is the best way to build a big following on social media? Big enough to get regular commissions and possibly start a patreon?

You'd probably have more luck with Yuri on Ice on tumblr

Fujoshis do a lot of commissions, from what I've heard

People always say this but I feel like it's wrong. Drawing fan art of popular shoes may help you retain a following but I don't think it'd be that helpful if you just started posting. If a show is popular than literally everyone is drawing fan art of it, ranging from absolute beginners to high level pros, you're piece has a very small chance of actually being seen. Now there is a chance that you do something cool and it picks up steam and you start getting a following that way but I think the odds are slim imo.

I think the best method would be to draw fan art of media that you actually enjoy and try to appeal to a niche audience. If for example you liked a video game that wasn't super popular, there's probably not that much fan art of it out there. That way if you do fan art for it, you're piece will stand out way more and you won't be forcing yourself to be dishonest and draw art of stuff you don't like.

Disclaimer: I haven't really tried either method, these are just my thoughts on the matter

I think when it comes to fanart, to be really popular, you need to be one of the few people that is creating content from the very beginning. That way, as time passes, more people will start to watch the show, get more involved in it, and look for art to fuel their fantasies; when that time comes, you'll be ready with artwork with all the pairings and shit that people like.
Pay attention to the new animes that are coming out in the next cycle and search around to find the manga or light novel to get a sense of what characters are popular and see the size of the Japanese audience it has. If it's fairly popular, it's probably going to be popular as an anime.
As for fujoshi, I'm not sure if you can rely on a lot of them for commissions as most of the audience is quite young. That's not to say the interest does not continue as they get older, but there is less of a chance that when the people who like BL now are old enough to have money they'll still be into it.
I spent quite a few years around that area of tumblr and I mistakenly assumed that the people I was following at the time were 18-20 as I was at the time, but actually, they were mostly 14-16 year old kids.

One site that I think is really getting quite a bit of artistic attention is instagram. It's the hip place for the kids to share artwork and even people with the shittiest art have about 5,000 followers (even without face pics). It's also a great way to showcase your artwork to normal people, however I feel the people who use the service have a very strong sense of "pastel goth"/ "kawaii" (or "unicorn poop" as I've heard it described as) in their artwork as that's what is trendy right now--and has been for about five years. You have to be willing to sort of fit in to get popular. That applies for tumblr as well.

What is the best way to build a big following on social media WITHOUT doing these three things?

Just do your thing, post it everywhere. If its worth a damn it will catch on. If not, back to the basement.
