Paint strokes on clear acrylic "glass" and photograph it. Photography tends to protect color integrity moreso than scanning. You can also use lighting to manipulate the brush strokes.

Photoshop allows for 1-bit brushes only.
Illustrator is a different story though, and you might be able to find those kind of brushes available for download or purchase somewhere.

Corel Painter is pretty sick for stuff like this.
Much better than photoshop and you can do mad things with watercolour layers and the brush range is great too. The interface sucks though.
