I have the define S and if you don't have a need for more than 3HDD's and 2 SSD's (or a mix) then the define S is better.
The only time the R5 is better is if you have a bay reservoir or need all the HDD slots.

I own the windowless version of both the Fractal Design R5 and Mini C. They are both whisper silent. The Mini C is in my bedroom which is a modest 14 x 10 foot bedroom. I cannot hear the pc at all even while sitting right next to it in the middle of the quiet night. My Mini C has four case fans — the two Fractal Design GP-12's that came with the fan plus two Noctua NF-F12's that I added. Inside the case is an i7-7700k, a Cryorig H7, an EVGA GTX 1080 Superclocked, and a Seasonic 650w SS-650KM psu. That's a lot of fans, and no noise at all eminating from my windowless Mini C.

Bonus — the cable management and overall modularity of both the R5 and Mini C are extraordinary.

Or Thermaltake F51 (without the tempered glass option) which is the same as the R5 with, I think, a smaller side fan aperture.

I can report its very quiet and generally solid and well put together.

No scientific view on air flow but my 7700k is running a 28 degrees, 1070 EVGA SCX3.0 (aka Firestarter Edition) is at 35 on a profile where I can't hear the case fans.

I didn't disconnect it, I made it so it doesn't shine out of the front, only the ring does. I also didn't plug in the HDD led. With only the ring being shining, its very possible to sleep in a dark room with it turned on.
