It will work op, but when you've booted up you'll need to run all the programs in /usr/bin once so theyre buffered into RAM. then you can safely remove the USB

No it wont work op. Obviously. If you insist on running an opperating system off USB then id recommend at least having a few USB 3.1 drives in raid 0 to house the OS

Slax will dump itself completely into RAM if you want, letting you remove the drive without reprecussion
