Autohotkey is really good for automation. Follow a shitload of people, wait a few days, in follow them, repeat ad nauseum, and you'll build an army no sweat. I have 10,000 followers on a Pokémon meme Twitter.

Most have APIs you can use. Twitter seems less aggressive about bot activity if you use iMacro instead though.
