Mysterio as a villain would probably end up with a lot of visual effects that would be too similar to some of the ones in Doctor Strange.
A different villain works in different stories. Mysterio is good for mindfucks and cool visuals. Venom is good for that visceral, to-the-bone kind of conflict, and Green Goblin is there for that very personal vendetta.
