That actually sounds like it could be really cool honestly. Aishi is following Jack on the ends of the Earth to kill him, who is following Aku to kill, and through her journey she meets the people Jack has helped throughout the show and slowly begins to realize that she was wrong about him.

I thought the wolf was more of a metaphor for Jack's will to live/ survival instinct, for the wolf during episode 2 it was kill or be killed and Jack meeting with it in episode 3 seemed to be him reconciling with this and his more "primal" side.
Throughout episode 3 Jack is nearly naked, has no guns or technology, almost looks like a caveman, and he fights the sisters in a forest in the middle of nowhere, far away from civilization.

In Japanese, one of her titles is "Ookami" or "Great Goddess"
