Is the 52 a cornerstone of the DC universe I should read?

Definitely yes. It's a good introduction to a lot of elements of the DCU and its superhero community apart from the major A-listers, and is also the point where the Multiverse gets reintroduced after being destroyed back during Crisis on Infinite Earths.
It's also a damn good story on its own merits and is fun to read along with even if you're new to the DCU.

Is Batwoman in it much or just a couple issues?
I've only read new 52 Batman and Batwoman so far and liked both, for the most part.

Batwoman isn't that important here and kinda sucks in it desu.
She was originally going to have an ongoing way way back when, like 2007ish, but the media hype and curiosity about a lesbian batcharacter made them suddenly cancel the plans for the ongoing, boot the creative off without telling them, and shove her in 52 real quick to get her out there. That caused her character to not be solidified and she basically plays second fiddle to Renee.

Renee Montoya is more of the focus character for that part of the story, but Batwoman plays an important role in it. She appears multiple times.

Which was pretty fantastic.

It is considered an event book and is actually widely loved
Which is super rare for these big organized corporate event stories.
And it is a great story for new readers cause while you may not know most if any of the characters, you'll be a fast learner and get a real feel just how much bigger DC is than just their usual main guys.
