I'd actually argue that Specialization is the worst, unless you get lucky and your specialty is something that lets you use all of the other nen types. Most Specialists seem to get even less choice in their nen ability than the other classes.
As for the best, I'd say Enhancer. Being really strong is always good for offense, defense, and speed, not to mention being able to heal and enhance other people and things. And that 80% in Emission and Transmutation just make it all the better.

It's not very specific, really. Conjurers really seem to be the ones capable to pulling off the most crap. Teleportation, for example, is an ability that for some reason is associated with Emmission, but a Conjurer like Novu can achieve what is basically teleportation with his portals. As long as it's somewhat limited by their chosen object, Conjurers can achieve any effect that they want. I don't know if that bullshit is limited to Conjurers, I don't know if an Emmitter couldn't make an ability that's basically a Nen blast, but when it hits you it makes you lose memories, for example.

If I were Manipulator, I would undoubtedly make an ability to control people, if only because it's the best use I think you can get from Manipulation, but I can't see many good uses for it, so I would probably go evil and most likely use it for control and rape girls. True, but from a stat standpoint, they only have one directly adjacent nen type on the chart, limiting their potential. The three on the top of the chart have much more potential. I'd rather be emission just because I'd have 80% in enhancement and manipulation. Emission seems so much more useful to me.
