Exactly. One of the biggest issues of CGI in Anime right now is the fucking 12FPS animation bullshit it has going on.

If you're using CRF, then no, don't waste your time on 2pass.
If you're using a straight bitrate tho, (vbr) then yes, it does help.

There is none, or ever will be.
It always depends on the content, then on the length.
You can have a 1:30 min long video with motion at all and you can make a 720p vidoe at very good quality EASILY.
But then there is a highly demanding video with lots of motion that you can barely fit into a 300p video

First of all I'm using webm4retards since I prefer the interface cuz it makes it quicker, and the more advanced commands are really not worth it for me personally.
I ALWAYS use CRF for the shit i post, starting off 30-40 (40-45 at the VERY worst) and change the resolution going from there.
Once I've reached something like 18CRF at 720p I just leave it there.
