If you're on a real bike sprinting up hills or on a track then you can build muscle but that won't translate to squatting under a bar it will make you better at cycling.
If you squat with the proper amount of volume (weight * reps) you don't even need isolation exercises after it and it can take as much time as you need to get the work in
Squats aren't an exercise you want to superset with anything. You get under the bar, survive the set and then sit down for 3 or 5 minutes maybe more if you're doing singles at 90%+ and then get back under the bar.
