I think I can solve that -- at least approximately.
I draw a black rectangle over her leg on a separate layer, then converted the image to grayscale.
The layered area matched the behind-the-skirt skin when the rectangle was 51% opaque.
So I made the area where the skirt only covers background black and 51% opaque.
Sliding variously colored objects beneath it seemed to give reasonably convincing results.

A really accurate answer would darken RGB in different proportions.
You'd need some non-black color at some other degree of transparency. If anyone feels up to the math;
Bare skin is 253,233,216 Covered skin is 168,121,151

Yes, you're thinking in the right direction. It's some shade of purple over her leg at least.
Wouldn't you need two differences to solve the equation though? Because take a white square for example: overlaying a grey square at 100% opacity would give the same end result as a black square at 50%.
If the artist actually used a transparent layer (which he probably didn't), we should compare the effect it has on skin to that on Megumin's cape. That's 82,66,106 for bare, 90,79,137 for covered.
Aaaaand it's unsolvable. The part over the cape makes it bluer, while the part over skin makes it purple/pink. I found a good match for either, but they didn't work on both.

We have 3 examples of covered vs. uncovered; Sunlit skin, shaded skin, and cape.
Which do not yield a definitive solution. But there'd be a least-squares "minimum error" answer.
That's why I did my check on a gray-scale image. That reduced the variables and a good deal of the subjectivity.

The problem was, if I set tolerance high enough to include boundary-pixels, a good deal of her hair was also selected.

I don't use Ps, but GIMP.
1. Select background with fuzzy select (magic wand).
1b. Select sharp corners with free select (lasso).
2. Grow selection by 1 or 2 pixels to get the edges.
(2b. Invert selection, feather, invert again. Not always needed.)
3. Color to alpha.
The color select tool gets a whole bunch of other stuff most of the time, so I rarely use it. Magic wand does the same thing with finer control.
