Since there's no way for us to exactly know what Hori thinks of its canonicity, you could take his words in that page as a confirmation or you could wait until the Vigilantes make an appearance

i'm on the canon camp myself the day trigger made an appearance, and it's the evidence is even stronger after Hori directly addressed Vigs in vol 13 (or 14?) and then Stain even made an appearance. But even in the face of all that, for some anon it's not a compelling enough evidence until the main series directly features the Vigilante trio (which, I think will appear once their story wraps up, since Hori isn't one to throw away interesting characters)

Yakuza are basically just mafia, their illegal actions edge more towards tax fraud, laundering money, real-estate, cooking books for politicians, ect.
They work hard to keep out of trouble, they don't just go around attacking and murdering people both because it's bad for business, bad for life, bad for their customers, and insures heat. They're not an immediate threat regardless of how well known they are.
