The number for genetic diversity is actually way lower. Usually in the hundreds, all the way down to just about 50 if the couples were specifically picked out of the existing population for their diversity.
The 10-40k thing was for simulations of interstellar travel that involved a bunch of factors which killed off large amounts of the population and wasn't specifically aiming at breeding for diversity but just normal relationships.
