I really do not see any single anime being the most influential.
Really what happens is a groundbreaking anime is made then subsequent anime build upon the groundwork that anime laid.
There have been quite a number of anime like this throughout the many decades, especially in the 70's and 80's when the medium began to enjoy mainstream popularity.
I would say that it is much harder to make an influential anime now than it would have been in the 80's.
That is just a broad statement about anime as a whole, now anime influences within genres is another matter.
If you are intensely familiar with a genre you can usually trace influences.
When I watch a show in some of the genres I enjoy I feel as though I have a clear idea of where the major influences and ideas originated.

Pokemon was not influential at all in the west, though it was in Japan. It was popular, yes, but it didn't really influence a whole lot. I would say DBZ was much more influential than it, since DBZ more or less introduced action to cartoons in the west.

you could argue Pokemon or DBZ, but I say Lupin III. Not only is it one of the most influential and important series in Japan but it's also responsible for influencing guys like John Lasseter and Steven Spielberg.

It's parodied and referenced it literally tons of anime/manga as well as american cartoons, disney films, and even video games.

Even things like Astroboy which despite being important to anime isn't really relevant anymore unlike Lupin which has new content every year. This year alone they came out with a hit film Directed by Koike himself.

If every member of the cast is wacky kooky krazy then none of them really stand out (barring some really masterful characterization). I don't know if it works in lupin or even if Goemon is actually foil (from what I've seen of the series he's just as goofy as the rest of them), but it's silly to discredit the idea of the writer/director deciding to try to change a character to enhance the rest of the movie, right? After all, isn't that exactly what Miyazaki did for Caligiostro?

I agree except I think it has 2 issues. Yes the music, and also because the original Japanese cast wasn't in it due to them using most of the budget on animation. Nothing can beat the chemistry japanese Lupin and Zenigata had. I'd argue that's a bigger flaw than the music itself, but regardless it's a great film.

I thought the they did alright, although I'm not a fan of the underhanded way in which the production dealt with the older cast. To give my opinions about each one:

Toshio Furukawa (Lupin): Good. I would have liked to hear him as Lupin again personally.

Banjou Ginga (Jigen): A great voice actor on his own but not really a fit for Jigen.

Mami Koyama (Fujiko): MUH DICK.

Kaneto Shiozawa (Goemon): Far too nasally IMO.

Seizo Kato (Zenigata): Serviceable, I suppose.

But there are many influential anime across the decades. He asked for THE most.
Otherwise, how can you not include, for instance, Mahou Tsukai Sally? It created a whole genre and spanned a lot of clones, the 70s and early 80s were plagued by their clones. Or Sailor Moon, which basically took the classic formula and subverted it.
What about Ashita no Joe's influence on sports? Or Candy Candy in shoujo? Or Versailles no Bara? Even if we talk only about science fiction and robots in general you can add Yamato, Cobra, Macross (yes, it's Yamato and Gundam mixed together, but it was extremely influential in its own decade) and Mazinger. In martial arts Ring ni Kakero, Hokuto no Ken and Saint Seiya were so influential that their influence spanned even to videogames with their weird techniques and use of spirit energy.
