It's built less as a collection of jokes, and more as a cohesive show tightly oriented around its themes of nostalgia, growing up, etc. (however you want to read it). The characters in K-On! feel more fleshed out, and despite being a pretty romanticized series, it does well to create a sense of realism. At this point obviously everyone knows Yamada's a good director, that doesn't really need to be restated. I don't like to diss Azumanga, because I really am a fan, but it never gave the impression of being anything more than "just" funny. Which isn't a bad thing! If that's all you're into and you think AD is funnier, then more power to you. But a lot of people, myself included, like to approach this stuff with a little more artistic seriousness, in which case there's really no comparing the two. I'm not really trying to convince you to change your opinions or anything, just explaining where I'm coming from.

I've watched Azumanga Daiou like 15 times and K-On like 6 times. The most significant difference—apart from the fact that they're completely different shows—is in the direction, animation and character designs. Modern KyoAni is all about animating the subtle facial shifts and mannerisms that convey what the characters are thinking and feeling. It makes them feel human and tells you a lot more about them. Your chances of empathising with a character skyrockets as they get less robotic and more human.

Have a look at just about any show from any other studio, and watch some characters deliver lines. In 95% of them, a characters face and body will be static throughout, only the mouth moving. Zero body language; pure autistry.
