Masterpiece in general is about mastery over the craft. Even boring or frivolous content can be turned into masterpiece by artist who has mastery over the media he/she uses.

In manga/anime there a lot of areas to express mastery in: art, storytelling, music, animation etc. If even one of these things are executed masterfully it can elevate the entire project to greatness and higher if multiple.
