There's the rub innit
In real life there's usually only three reasons why it happens.
1. The most common one - they are at least one of not attractive, not agreeable, or not unburdened (i.e. they have some serious fucking baggage, emotional or otherwise).
2. They legit just don't want to get married or at least aren't in a rush. Remember that if you're going for a doctorate you'll be at least 25+ by the time you finish. In fact plenty of people are approaching 28-30 by the time they get that degree.
3. They HAD a boyfriend and got dumped.

In anime it doesn't really work that way, for the most part, because the entire point of having the cake is for her to be attractive.
