They were initially drawn to each other because they both have an innate understanding of what others are thinking, and have a penchant for manipulating others resulting in their mutual interesting in Tomo and Jun's relationship. They have a certain kinship in that regard.

Misuzu has an additional interest in Carol because of Misuzu's larger motivation to become more open and affectionate with others. She sees the way Carol is so casually affectionate with Tomo and envies that, and hopes to emulate it. She has great disdain for Carol's cloying demeanor, but aspires to be more like her just the same.

Carol, on the other hand, hopes that Misuzu can help her break through to Sempai, but that plot hasn't gone anywhere in probably almost two hundred chapters now.

So there you have it. They're often outwardly antagonistic but have a mutual respect and camaraderie. They're mismatched friends, a manga odd couple.

The problem is that Jun's biggest weakness are big ol titties, which is why he avoided watching Carol's carols when she jumped rope and why he looked away when she walked out with a bikini. His best friend having big ol titties only makes his physical attraction harder to hold back.

I think it's simpler than that. Jun admired the Tomo of old and wanted to surpass her, perhaps as a show of gratitude for setting him down on the path of manliness. Just like those homolust rivalries that pop out every so often, with one participant in the relationship vowing to surpass the other while respecting the other's strength greatly.

Tomo becoming more feminine conflicts with his desire to surpass her. If she starts straying away from all the past stuff she used to do, Jun loses his chance to surpass her. So he can't acknowledge her as a woman, at least not yet. So either he has to beat her first before he can think of her as a woman, or he learns that, even if Tomo wants to become girlier, that doesn't mean she'll lost all her tomboy traits and strengths. This is what I think, at least.
