After around 2002 or so, it seems like the definition of harem changed to only refer to shows that had a shitty audience-insert protagonist. So no.

People describe harem as any show with a male main character and many love interests, but good shows that have an interesting, charming main character with many love interests and decent writing surrounding their relationships are generally not referred to as harem.

If he was a supporting character who appeared several times as a background character before his debut, would you suspect that he was going to become more important later on, due to his very distinctive and well developed design, befitting of a protagonist? Does his usual expression portray personality beyond "preachy goody two shoes", "passive aggressive Japanese high school student", or "/b/tard"?

I can think of a few exceptions (Konosuba, for example), but these are some good guidelines that will serve you well most of the time. It's not hard to tell when the author actually intended for you to remember the protagonist.
