Could Grisha have experimented on his own son with small doses way before he actually injected the full payload into him? And even if he did, he must have had obtained some of Frieda/Uri spinal fluid if he were to give Eren any semblance of the First King's abilities. How did he do this/does this even make any sense?

100 years ago, the First King's will was to create a world of peace between Titans and Humans but this never actually brought peace. Those who inherited the memories of humanity were driven to madness when faced with the fact that they have to live the life of a Titan and a Human as a shifter. The people within the walls are bred to hate and fear Titans despite the Reiss family's/First King's memory manipulation, and the people who inherit the First King's power and will are no different. When faced with the decision to turn their backs on everything they have known and wipe out the Titans with their new power, they are unable to continue hating themselves enough (Titans) to do so and instead become like Frieda who has bouts of insanity due to loss of identity (similar to Reiner and Bertholdt). This means that there cannot be peaceful coexistence between them.

This is where Eren comes in, someone who is observed as one who embodies an indestructible will. Eren and the First King shared this quality with the creator of the Titans, but it will be Eren who finally brings about change in the world.

For all we know, humans and Titans may very well have coexisted for 2000 years when the God created them due to technology (which has now been lost). Humanity doesn't remember this though because of the First King and their isolation from the outside world via the wall. It could be possible that there has been an ongoing 2000 year war between shifters who wanted to renounce their humanity and turn the world into the dominion of the Titans (Beast Titan) and shifters who wanted coexistence (First King).
