Okay so I got a low res texture, what would I want to do to upscale it and have it not look just blurry and crappy.

This will not clear up anything for you. Work will actually be needed. You will need to adjust or re-paint whatever texture you are trying to high res. Substance lets you export out to 512 all the way to 4096. That does not mean it will take a low res texture and high res it.You can take a high res and make it low res. It does not magically do the work for you.

Is this any good for hand painting, I got Quxiel as it was on sale recently and i'm happy about buying it.

But i'm told painter is better for hand painted textures.

True.
Since you can use the regular Photoshop brushes you have much more options than in Painter.
But for traditional handpainting in 2D i prefer Krita over Photoshop.

Here's a thing in Substance painter, it's pretty easy to get the hang of, and you can get decent looking thing quickly.
