The block is simply using the shield as a shield (duh), it reduces damage taken from the front and slows you down a bit (shields with better blocking slow you down more), this consumes charge ("ammo" for the shield, like any other weapon).
The reflect instead create a small "projectile" that reflect all enemy projectiles (IIRC the only exceptions are laser beams and Retarius webs), this consumes some charge everytime it's used.
You can use both functions but their effectiveness varies with the shield.

Example: the reflector has shit blocking and low charge but is fairly effective in using the charge for reflection and the reflect comes out instantaneously. The tower shield is the complete opposite, big, lot of charge, great blocking radius and damage reduction (180°) but both slows you down when it's out and the reflect takes a good 1-1.5 seconds to activate.
