The audio isn't fake, though the context is dubious. In that we don't know how the audio was supposed to have been used (it might have even been a joke left in the game.) It could very well have been a gay event, or something else. It does sound hella gay at least.
There's reasons other than being in the closet that people might be homophobic. In high school, I wasn't religious, I didn't come from a family with a lot of homophobes, and for good or for ill I eventually turned out straight, but I was absolutely convinced that The Gays were a near-supernatural force of evil summoned into the world by a secret cabal of environmentalists who wanted to reduce the overall human population by discouraging breeding and then force what was left after a few generations to submit to a primitive hunter-gatherer lifestyle, and that because I was onto their evil scheme, they were out to ruin my life and were thus behind my failure to get laid and the way every girl I developed any kind of feelings for turned into a drug addict with an abusive boyfriend. So I always figured that, or at least the basic sentiment behind that conspiracy theory that gay people are different from the rest of us and there was something not right about someone who was different in that way, was underlying Yosuke's homophobia.
