Super Mario Odyssey spoilers:
Believe me if you want to, or not. Look away if you want to be surprised.
The sub-point of this game is Mario exploring the planet he lives on. He's already been to space, but as far as we know, we've only seen the realms of the Mushroom Kingdom and Bowser's Kingdom. This game changes that, as we will get to fly around the world.
Bowser kidnaps Peach as always, Mario goes to rescue her, but this time he fails. Mario gets a "Game Over" and "dies" (sort of). He stomps on Mario's hat to signify his victory.
Bowser, believing that he's defeated Mario for good, decides to marry Peach. Peach is shocked and heartbroken as she also believes Mario to be dead.
Meanwhile in "limbo", that's the Halloween Town looking-esque stage (it serves as the tutorial stage), Mario meets Cappy. Cappy is a ghost, this is why he's able to possess things and grants Mario the same ability. He decides to help Mario and brings him back to life "humorously by giving him a 1-up." He also chooses to go with Mario, because he has his own goals, as some other "hat ghosts" are causing a fuss. He possesses Mario's hat, and the quest begins from there.
Luigi is also playable and has the exact same stages, but a different story.
Luigi, also believing his brother to be dead, realizes that he must step up and become "Super Luigi". He rushes off to save Peach, because that's what a hero does and is what his brother would want." Before his journey technically begins, he also gets a "Game Over" and meets his own hat ghost. His adventure starts from there. Luigi plays as you would expect him to play. Longer, higher, floaty jumps, but slower running speed and poor traction.
There are a fuckton of cameos and references to other Mario game in Odyssey, be on the look out.

As far as the amiibos go:Any Mario amiibo simply gives Mario coins and sometimes a costume, depending on which amiibo it is.
Luigi amiibo unlocks Luigi right at the start, when you'd normally have to wait until near the end of the game to get him.
Peach restores your health, and gives you 1-ups.
Rosalina swaps Peach out for Rosalina (instead, you're saving her). The model and voice change. Any in-game references also change to Rosalina.
Yoshi spawns a yoshi in front of you (if you can have a yoshi in that stage).
Bowser makes the game more challenging.
Wario and Waluigi give you outfits that color correspond to them.
Not sure what the rest do.
