First decide on a couple gimmicks that make your world different from Earth. Such as more/less water or whatever.
The create a bunch of ecosystems, some are similar to Earth but not all have exact analogues, such as a forest of sessile invertebrates instead of plants.
Although it's possible that alien life won't resemble Earth life at all, I suspect that their will still be some convergent evolution. So you can either create aliens by combining Earth life until you end up with suitably exotic chimeras that pass as aliens.
It depends if you want unique looking lifeforms or more of the Star Wars kind that look like Earth life.
Create a bunch of creatures first, then pick one to advance into an intelligent lifeform (though it doesn't need to necessarily be a tool user).
Cultures tend to directly affected their environment, with the most warlike cultures developing in areas with the least resources.

Creatures like kaiju can't technically exist on Earth because they would collapse upon themselves unless they were made of exotic matter.
Giant creatures exist more easily in liquids because the water removes a lot of the stress that affects large animals.
Super huge beings could exist on other planets, but they would not be Earthlike, they'd be very different.

Basically whenever I got involved in some Harry Potter fan creative thing and it fell to me to make a BBEG, my default villain was a Darklord who wanted to expose magic to the world and create a new age of heroes and villains.
