I doubt it, not with .22lr and not with a tank like 452. The only thing I can see mattering is making sure you indexed the barrel to the exact same place as before it came off and neither overclocked nor underclocked it.

My dad worked a job that sent him away from months, when he came home for a week he would sleep for half of the week then reload. I really wanted to spend time with my dad so I sat and watched I asked alot of questions and he answered everyone. He realized I liked guns he bought me a 22-250 for my first ever gun at age 10 (still own it). I procured alot more firearms when I met my wife, we went through alot of hard times and thought maybe i should give up my shooting and guns. So one night i told my wife I'm selling my guns to help pay for shit. She said fuck you if you sell one goddamn gun ill kick your ass. Now Im not poor and have 17 guns in my collection. So long story short life inspired my gun addiction.

Hey /k/, have shot my newish Rough Rider quite a few times, and I'm enjoying it, but .22LR is obviously pretty underwhelming, especially out of such a large handgun. Is paying the 45$ for a .22WMR cylinder worth it?

You could try it out I suppose, but honestly you'd be better off saving up for a new gun.
If you want something with more oomph, and you like single-actions, a Ruger Blackhawk is pretty good, there's even Convertible models, like pic related, a .357 Magnum revolver that comes with an extra cylinder for 9mm Luger, giving you the option of very cheap plinking with something with a bit more authority to it.

Assume you are involved in a justified self-defense situation where you shoot someone and have stopped the threat but did not kill them. Are there situations where Good Samaritan laws would compel you to administer first-aid to the suspect while waiting for the police/ambulance? Would having some medical training (ex. being a licensed doctor/nurse/EMT) change anything? Would "stand your ground" laws overrule Good Sam laws?

I'm not a lawyer, so don't take a thing I say as law or fact, but personally I would not being that you have perfect reason to believe the suspect is still a threat to your well being. You retreat and call for help.
