What they're doing is introducing some molecules that bind to proteins and other structures of interest whilst leaving lipids alone, that then polymerize into a gel when they heat it which fixes everything that's bound to it, allowing them to remove a lot of the stuff that prevents imaging from being easy (like lipids) by washing some charged molecules through the gel.
